Cytoprotective effect of alpha-2-macroglobulin against pesticide-induced generation of ROS in neuronal SH-SY5Y cells

Introduction: Many clinical studies have demonstrated that continuous exposure to pesticides, especially organophosphates and pyrethroids, causes toxicities such as carcinogenicity neurotoxicity lead disorders diabetes, lung cancer, neurodegenerative diseases. The mechanism underlying pesticide-induced involves the production of ROS, which neuronal injury through oxidative stress. Methods: In present study, SHSY5Y cell line was used investigate effect pesticides chlorpyrifos (organophosphate), aldicarb (carbamate), deltamethrin (pyrethroid) on ROS-mediated toxicity protective alpha-2-macroglobulin (a2M), a protease inhibitor beta-amyloid plaque scavenger in human brain. For viability cytotoxicity, MTT assay performed. To monitor ROS production, assays DCFHDA, H2O2, MDA were performed, along with activity antioxidant enzymes superoxide dismutase glutathione peroxidase. Results: experimental findings suggest cytoprotective role for a2M humans. Conclusion: Hence, could be possibly agent against caused by pesticides.
